Mouse Monoclonal Antibody
A monoclonal antibody is a type of antibody produced from a single cell known as a hybridoma that has a single specificity and is of one class or subclass. The production of monoclonal antibody begins with the injection of a mouse with a particular antigen phases. In a few weeks, when the B lymphocytes begin proliferating in response to the antigen, the spleen, a primary source of these lymphocytes, is removed from the mouse and fused in vitro with myeloma cells by adding polyethylene glycol. The hybridoma clones are formed through a three-step process: selection, screening, and cloning. The hybridoma cells are selected by cultivating in HAT supplemented media and screened by ELISA to identify those hybridomas that are producing antibody molecules of the desired specificity. The hybridoma clones can be used to produce a particular antibody in mice (ascities fluid) or tissue culture medium. The total time required to produce a monoclonal antibody in Genesis Biotech Inc. is about 120-180 days.
Advantage of Mouse Monoclonal Antibody
Advantages of mouse monoclonal antibody are: 1) specificity. Each one recognizes only one epitope of the immunogen, 2) Immortality, 3) Mice are small so they require less immunogen than rabbits to elicit immune response.

Scope of Work

2. Immunize 3 mice or 1 rabbit. ˇ@
3. Check immune response and report to customer. 30% Charged
4. Take spleen and isolate polyclonal antiserum. ˇ@
5. Conduct cell fusion. ˇ@
6. Conduct ELISA screen of hybridoma clones. ˇ@
7. Send supernatant of up to 20 positive hybridoma clones and antiserum for each project to client for additional assays. 30% Charged
8. Conduct additional assays and select 3 hybridoma candidates.(Alternatively, Genesis perform this step under customer's agreement )

9. Sub-clone the selected hybridoma candidates. Screen sub-clones by ELISA and select 3 hybridoma-mAb sub-clones from each candidate. ˇ@
10. Send supernatant of hybridoma mAb sub-clones to customer for additional assays. ˇ@
11. Conduct additional assays and select at most 3 hybridoma mAb sub-clones for each project (Alternatively, Genesis perform this step under customer's agreement ). Additional sub-clones can be purchased with additional fee. ˇ@
12. Send the selected hybridoma sub-clones to customer, or produce monoclonal antibodies at Genesis Biotech Inc. for an additional fee upon customer's request. ˇ@ 40% Charged
